CTOnews.com, November 25 (Xinhua)-- scientists have recently developed a new algorithm that has successfully discovered 188 rare and previously unknown CRISPR connecting gene modules in billions of protein sequences, providing new insights into the use of CRISPR systems and understanding the functional diversity of microbial proteins.
Photo Source: Carlos Clarivan / Science Photo Library the project is led by Professor Feng Zhang, a pioneer in CRISPR technology, with team members from the McGovern Institute of brain Research at MIT, the Broder Institute at MIT and Harvard University, and the National Center for Biotechnology Information (NCBI) at the National Institutes of Health. The results were published in the journal Science on November 23.
The team developed a new search algorithm-based on Fast locally sensitive Hash clustering algorithm (FLSHclust). Using this algorithm to mine three major public databases, 188 new CRISPR systems were identified, and four of them were characterized in detail.
These new systems may be used to edit mammalian cells with less miss effect than current CRISPR-Cas9 systems, or they may be used for diagnosis or to record cell activity.
This study found several new variants of the known type I CRISPR system, which can be used to develop more accurate gene editing techniques in the future.
In addition, the researchers also found some new mechanisms of IV-type CRISPR systems, as well as VII-type systems that precisely target RNA (which may be used for RNA editing). The researchers say the study highlights the unprecedented diversity and flexibility of CRISPR systems.
